I'm going to log my journey in tissue culture here. Feel free to post anything relevant, ask questions, or share answers. Once I consider myself competent in various aspects of TC, I'll add documentation and protocols to a separate tissue culture thread in a Knowledge Forum.
 
Start-up costs. Far and away the MOST expensive piece of equipment is the Laminar Flow Hood. You can use a LARGE pressure cooker instead of an autoclave for about $100. You don't need a microscope unless you're doing apical meristem TC, in which case you need $100-$200. Everything else is not particularly expensive and can be purchased on Amazon (generic lab glass from China, misc. lab equipment) and PlantCellTech.com (specialized TC store that makes a bunch of cool videos and sells high-quality stuff).
 
I had a lot of deferred maintenance I needed to take care of in the garden. I'll be back on the TC tomorrow or Saturday. I'm going to make some new medium (rooting) and fill about 10 16oz "deli containers" and 21 test tubes (I'll take a close-up pic of those bad boys - got them at PlantCellTech and I'm totally geeking out on these gigantic tubes. I'll probably take nodes from a few mothers and I'm going to use the test tubes to germinate some seeds. Side note - I will NEVER germinate another seed outside of tissue culture. There are so many benefits - each pheno "mother" plant is in a sterile jar replicating, taking up virtually no space and not requiring frequent watering/nutrients/IPM.
 
I'm autoclaving a new batch of medium for multiplication. Once it cools off I'm going to put them into some deli style containers for nodal explant TC and into some test tubes for seed germination. I don't foresee ever popping another seed outside of Tissue Culture as the advantages of keeping a clean mother in a small container right from the beginning are huge. I don't think any of the containers are going to require more attention than every couple of months unless I choose to give them more. Culling phenos will just be a matter of pulling a small container off a shelf and cleaning it.
 
My vision is not what it used to be but it LOOKS like my first effort at TC has NOT gotten contaminated and is starting to show some roots. I used a container that was too small so I'm probably going to move them into a deli container
 
I don't THINK there's supposed to be condensation in the containers. The medium was very warm when I poured it in and I probably put the lid on too tight. Hopefully no long term consequences as I'm moving it today.
